The goal of this work was to develop, optimize, and validate an automated, digital-image-analysis-based microscopic scanning system for the detection of immunofluorescent-antibody (IFA) stained Cryptosporidium sp. oocysts on well slides and/or filtration membranes. Automated counting versus manual counting trials were carried out utilizing IFA-stained preparations with various amounts of interfering detrital material present. We also sought to optimize staining, mounting, and optical properties of water concentrate preparations using the instruments' quantitative capabilities. Automation of microscopic scanning of filtration membranes or well-slides for Cryptosporidium sp. oocysts will reduce assay subjectivity, and analytical costs as skilled microscopist time is minimized, and more of the water concentrate can be practically analyzed. In addition, analytical techniques which require microscopy and epifluorescence staining can be quantitatively optimized and compared.
